The active component of
pyrazinamide, pyrazinoic acid had been synthesized in 1934, but its
activity against M. tuberculosis was then not recognized.
Pyrazinamide is active only against M. tuberculosis. It
requires an acid milieu for its activity. It reduces the risk of
relapse and has allowed shortening the duration of chemotherapy when added
during the first two months of treatment.
